Sequencing savings with TempO‑Seq

Fewer reads per sample changes the entire cost equation

The most important sequencing advantage of TempO‑Seq is read depth. The S1500+ panel can be planned from about 0.67 million reads per sample, while TempO‑Seq Whole Transcriptome is typically planned at about 3–6 million reads per sample. Standard RNA‑Seq projects are commonly planned at 20–50 million reads per sample. That difference determines how many flow cells you need and how much sequencing costs.

The core argument

Same sample count. Very different sequencing burden

For 96 samples, 20 million RNA‑Seq reads per sample requires 1.92 billion reads. On a 400 million read flow cell, that is five flow cells. A 96-sample S1500+ project at 0.67 million reads per sample needs only about 64 million reads.

Read depth first

Start with how many reads the biology actually needs

Rather than centering the page on a single sequencer, this version starts with the assay. Once the read requirement is set, the number of flow cells and the sequencing spend fall out of the math automatically.

0.67M

S1500+ panel

The lowest read-depth option on this page. At 96 samples, the project needs about 64.3 million reads before any planning buffer, which can fit comfortably on a small Illumina flow cell.

3–6M

Whole Transcriptome

Even the broad TempO‑Seq Whole Transcriptome workflow is planned at a fraction of the reads commonly used for standard RNA‑Seq. The calculator lets visitors move anywhere within the 3–6 million range.

20–50M

Standard RNA‑Seq

This is where the cost gap opens. More reads per sample mean more flow cells, and standard RNA‑Seq also adds library preparation plus RNA purification before sequencing begins.

Toggle the panel, read depth and Illumina flow cell

This calculator mirrors the argument in your email: compare the lower read requirement of TempO‑Seq with a conventional Illumina RNA‑Seq workflow that also carries RNA library-prep and purification costs. Choose among several smaller Illumina flow-cell configurations to see how the economics change.

Planning illustration only. The TempO‑Seq side compares sequencing burden and does not include the TempO‑Seq assay-kit quote. The RNA‑Seq side intentionally includes library prep plus $15/sample RNA purification because those are additional conventional workflow costs highlighted in your email. Flow-cell pricing uses a public Illumina US list-price snapshot revised May 31, 2024. Actual pricing, discounts, read structure, utilization, output and project design may differ.

The 96-sample argument

The email logic becomes very easy to see

With the default 96-sample comparison, a 400 million read flow cell makes the point immediately. The TempO‑Seq project fits on one flow cell, while a 20 million read RNA‑Seq project needs five. The conventional workflow then adds RNA library prep and purification on top.

96 samples with S1500+

At 0.67 million reads per sample, 96 samples require approximately 64.3 million reads. That is well below the 400 million read capacity used in the original NextSeq example.

64.3Mtotal reads

96 samples with standard RNA‑Seq

At just 20 million reads per sample, the same 96 samples require 1.92 billion reads. Dividing that by a 400 million read flow-cell capacity gives 4.8, which means five flow cells before considering any utilization buffer.

Multiple public list-price comparisons, not one cherry-picked platform

The calculator includes several Illumina flow-cell families so a visitor can test the same read-depth argument across different throughput levels. The shorter-cycle and longer-cycle prices below are paired within the same platform family to reflect the cost logic in your email.

RNA‑Seq cost layers

Sequencing is only part of the conventional RNA‑Seq bill

The email argument is strongest when the page shows all three conventional cost layers together: library prep, RNA purification and sequencing. The 96-sample example below keeps those costs visible instead of comparing flow-cell prices in isolation.

Workflow 96-sample library-prep example RNA purification 20M reads/sample on 400M flow cells Flow cells required Why it matters
Illumina Stranded mRNA comparison $3,446.80 $1,440 1.92B total reads 5 The cheaper RNA library-prep example still carries purification and five 400M sequencing runs at 20M reads/sample.
Illumina Stranded Total RNA comparison $7,364.72 $1,440 1.92B total reads 5 The higher library-prep spend makes the total cost gap even larger before sequencing depth increases to 30M or 50M.
The two library-prep values above are the 96-sample example costs supplied in your BioSpyder email context. The calculator scales them proportionally for illustration. They are not represented as live Illumina account pricing.
